چکیده

Basophil activation is a key finding in allergic reactions and also observed quite frequently in infectious diseases and autoimmune disorders. In allergic reactions, basophil-derived mediators such as histamine, contribute essentially to clinical symptoms. During IgE-dependent degranulation of basophils, a number of cell surface membrane and cytoplasmic molecules become activated, show altered expression, or are translocated into the cell surface. Although little is known so far about the exact role of these activation-linked cell surface antigens, several of them are employed as diagnostic parameters in allergic disorders. Other molecules are involved in the process of signalling and the consecutive release of pro-allergic mediators, and have therefore been proposed as potential targets of therapy. The current article provides a summary on activation-linked cell surface and cytoplasmic antigens in basophils, with special reference to potential mechanisms underlying re-translocation or over-expression in activated cells, relevant signalling pathways, and clini-

The basophil activation test by flow cytometry: recent developments in clinical studies, standardization and emerging perspectives

The diagnosis of immediate allergy is mainly based upon an evocative clinical history, positive skin tests (gold standard) and, if available, detection of specific IgE. In some complicated cases, functional in vitro tests are necessary. The general concept of those tests is to mimic in vitro the contact between allergens and circulating basophils. The first approach to basophil functional respo...

Expression of Basophil Activation Markers in Pediatric Asthma

Background: Asthma is very common in children and its diagnosis is based on clinical manifestations, which can be misdiagnosed as other respiratory diseases with similar signs and symptoms. Objective: To analyze the expression of ST2L and CD203c in the diagnosis of pediatric asthma. Methods: Basophils were purified from whole blood samples of patients and healthy controls using Ficol-Paque grad...

Commentary: The Expression of CD123 Can Decrease with Basophil Activation: Implications for the Gating Strategy of the Basophil Activation Test

BACKGROUND Basophil activation test (BAT) reproduces IgE-mediated allergic reactions in vitro and has been used as a diagnostic test. Different markers can be used to identify basophils in whole blood and have implications for the outcome of the test. We aimed to assess changes in the expression of CD123 and HLA-DR following basophil activation and to select the best gating strategy for BAT usi...

Differential response of human basophil activation markers: a multi-parameter flow cytometry approach

BACKGROUND Basophils are circulating cells involved in hypersensitivity reactions and allergy but many aspects of their activation, including the sensitivity to external triggering factors and the molecular aspects of cell responses, are still to be focused. In this context, polychromatic flow cytometry (PFC) is a proper tool to investigate basophil function, as it allows to distinguish the exp...

The basophil activation test reflects the severity and the threshold of allergic reactions to peanut – a double-blind-placebo-controlled peanut challenge study

Results 44 peanut allergic children (median age 5 years) reacted to peanut on DBPCPC with clinical symptoms than ranged from oral allergy syndrome to anaphylaxis. 61% of patients reacted to 0.1g of peanut protein. The mean %CD63+ basophil at 10 and 100 ng/ml of PE was independently associated with severity (p=0.012) whilst CD-sens (1/EC50x100) was independently associated with threshold (p=0.03...
